Abstract
Continuous culture devices that permit different removal rates for solids and liquids were used to evaluate effects of solids retention times of 16, 23, 28 and 40 h on microbial metabolism. Triplicate fermentations of 8 to 10 d duration were conducted at a liquid dilution rate of 6.25%/h. The substrate was a semi-purified diet with zein as the major protein source. Digestion coefficients for zein were 32.62, 54.77, 63.62 and 77.49% with solids retention times of 16, 23, 28 and 40 h, respectively, while the corresponding ammonia concentrations were 1.3, 8.9, 12.7 and 19.5 mg/100 ml. The amount of N incorporated into microbial cells did not increase with increased zein digestion and available N. Dry matter and acid detergent fiber digestibilities increased as retention time increased, but tended to plateau at 28 to 40 h. Neither total VFA production nor molar proportions appeared to be affected by solids flow rate. While extension of the solids retention time to 40 h increased the extent of digestion of dry matter, acid detergent fiber and protein, it had little effect on the efficiency of fermentation as measured by VFA production or microbial protein production per unit of dry matter digested.
