Abstract
Respiration and motility of washed porcine spermatozoa in the presence of bovine follicular fluid were studied. A Gilson Differential Respirometer was used to measure respiration rates (ZO2 values). Final motility scores were visually estimated using a microscope.
Whole bovine follicular fluid (WBFF) as 5, 10 or 20% of the suspension medium had a detrimental effect (P< .05) on respiration rates and motility as well as caused head-to-head agglutination relative to spermatozoa suspended entirely in Ca++ -free Krebs-Ringer phosphate buffer (KRPG) containing 1 mg glucose per milliliter. Dialyzed (DBFF) or heated (30 min at 60 C) bovine follicular fluid (HBFF) was less toxic to spermatozoa than WBFF. Spermatozoa suspended in 20% HBFF exhibited significantly higher (P< .05) respiration rates than KRPG suspended controls.
