Abstract
The functioning of oxidative phosphorylation in porcine skeletal musculature and its relationship with postmortem muscle quality was investigated. Mitochondria were isolated from biopsy samples obtained surgically from biceps femoris muscle of 20 Dutch-Landrace and 20 Pietrain pigs under general anaesthesia, and their respiratory rate and respiratory control ratio (RCR) measured polarographically. Two to 4 weeks post-biopsy, the animals were slaughtered and muscle quality was determined.
Average muscle quality was significantly lower for all characteristics in the Pietrain breed, and the mitochondria isolated from the pigs of this breed showed a lower mean RCR as compared with the Dutch-Landrace pigs. This lower RCR was not due to uncoupling of oxidative phosphorylation, since addition of oligomycin strongly inhibited respiration in all preparations, but was caused by an inhibited respiratory rate during the period of measurement in which the mitochondria were phosphorylating actively. Since this inhibition could almost completely be abolished by uncoupling these preparations artificially, it was concluded that a decreased capacity of the phosphorylating system was present in these preparations.
