Effect of Some Buffers on Storage and Freezing of Boar Spermatozoa

Abstract

Seven zwitterionic buffers titrated with Tris and three combinations of sodium and potassium hydroxide to pH 7, along with Tris-HCl, sodium citrate and sodium-potassium phosphate of the same Ph, were investigated for use as extenders for boar spermatozoa. Their suitability was measured by percentage of motile spermatozoa and by the release of glutamic oxaloacetic transaminase (GOT) from the spermatozoa. In general, the zwitterionic buffers maintained a higher percentage of motile spermatozoa than did the other three buffers but caused a greater release of GOT from the cells. When combined with the zwitterionic buffers, Tris was the titration base which best maintained sperm motility, but this combination also caused the highest GOT release. HEPES and TES were the superior buffers over all treatments as judged from motility and GOT release. There was a significant negative correlation between motility and extracellular GOT in samples stored at 5 C. Dilution increased extracellular GOT values.
