Distribution and Excretion of 14C-Labeled Polydiethylstilbestrol Phosphate in a Steer1

Abstract

A steer was injected with 767, µc. of 14C labeled polydiethylstilbestrol phosphate (PSP), and the accumulation of radioactivity in liver, muscle, fat and blood of the anabolic agent was measured at several times during 125 days. Feces and urine were collected and analyzed during the trial. At slaughter, radioactivity in various tissues was analyzed and uterotropic activity in excreta, blood and tissues collected at biopsy and at autopsy was determined in immature mice. Radioactivity was highest at 5 days in liver and was 31.6% of the injected dose. This level of radioactivity declined slowly and 7.6% was present at slaughter. Blood had radioactivity throughout the trial but no radioactivity was detected in fat or muscle at any time. Radioactivity recovered from urine and feces was 25.4 and 79.7% of the total dose, respectively. Radioactivity in small amounts was found in spleen, kidney, lungs and heart and the total radioactivity recovered from tissues at terminus was 10.5% of the administered dose. The uterotropic activity of feces and urine was 7 to 18% of the theoretical amount of DES based on radioactivity. PSP was not uterotropic. No uterotropic activity was in blood, liver, spleen, or kidney, and the injection site was weakly uterotropic, having less than 0.1% of that expected from its radioactivity.
