Mammalian Spermatozoan Metabolism in the Presence of Thalidomide

Abstract

The toxicity of thalidomide and phthalic acid to mammalian spermatozoa was tested using bovine spermatozoa both in the absence and presence of exogenous substrates and seminal materials. The toxicity of phthalic acid was as great as that of thalidomide; the presence of high levels of either compound resulted in decreased respiration of the spermatozoa. Thalidomide significantly (P<0.01) depressed respiration in the presence of fructose, pyruvate and succinate, but had little effect on the motility of the spermatozoa during the 37° C, 4-hr, incubation period. CO2 production under anaerobic conditions was not altered with treatment. These results suggest possible mechanisms whereby thalidomide may exert its teratogenic effects.
