Abstract
The purpose of this experiment was to evaluate quantitatively the possible changes in free amino acids or ninhydrin reactive substances (NRS) in the plasma of sheep subjected to captive-bolt or electrical preslaughter stunning. Thin-layer chromatography and automatic amino acid analysis were used to confirm and quantitate the plasma responses. Thin-layer chromatography gave nine distinct spots or groups of spots from plasma samples. These spots or groups of spots were tentatively identified by color and R( value. Densitometric scanning indicated that the relative concentration of isoleucine decreased (P<.05) and the concentration of valine increased (P<.01) with electrical stunning. Plasma amino acid nitrogen was not changed significantly. Automatic amino acid analysis indicated that seventeen amino acids and thirteen NRS were resolved from sheep plasma. The total concentration of amino acids increased (P<.05) when electrical stunning was used. The increase in the plasma level of valine resulting from electrical stunning was confirmed by both methods. Some metabolic aspects of this stunning effect in sheep are discussed.
